N-acetyl-D-glucosamine Assay
N-acetyl-D-glucosamine (GlcNAc) is phosphorylated by the enzyme GlcNAc Kinase (NagK) and ATP to form GlcNAc-6-phosphate and ADP. Using a coupled reaction in the presence of NADH, the amount of NAD formed is stoichiometric with the amount of GlcNAc, and is measured by a decrease in absorbance at 340 nm.
Calibrator curves of GlcNAc using a kinetic assay performed in a microplate (left panel) or cuvette (right panel). The assay was performed as per the method described in the applications manual.
